That’s mold/biofilm.
I’ll save you the time. Not trying to be discouraging but as a horticulturalist with a love of spruce and all sorts of gynnosperms, you’re not going to be able to root that in water.
You need a potent rooting hormone, soil, and time . Picea and the like are not props that just root like houseplants. They need to be snipped at the right time of year and really are a bit more advanced in terms of getting to root. Do some research on the best propagation methods for them to get an idea of what it entails. It’s a challenge but very rewarding if successful!
